Traits génétiques complexes.
Mon laboratoire s'intéresse à des traits génétiques complexes et utilise des techniques de
biologie moléculaire et biologie computationelle.
La pression artérielle et l'hypertension sont parmi les intérêts principaux de ce laboratoire. L'hypertension est quantitativement le facteur de risque cardiovasculaire principal. Des estimations d'hérédité indiquent que 30-60% de ce risque est génétique. Mais la pathogénèse de l'hypertension artérielle n'est pas claire. Nous travaillons avec plusieurs cohortes Européens et Américains afin de mieux connaître la pathogénèse de l'hypertension artérielle essentielle. Le laboratoire collabore actuellement dans un effort de consortium d'investigation de la pression artérielle par association « genome-wide » (CHARGE et ICBP) et nous avons effectué une analyse de ~70'000 individus qui ont tous été génotypés pour des centaines de milliers de marqueurs. Les résultats indiquent que la contribution génétique à l'hypertension est constituée d'une multitude de variantes avec faible effet (pour des variantes avec une fréquence de >5%) Un autre intérêt principal de mon groupe sont des efforts de séquençage à large échelle pour découvrir les déterminantes génétiques cardiovasculaires dans un spectre d'allèles plus rares. En utilisant les données du projet « 1000 Genome Project », ce laboratoire est impliqué dans l'analyse des données avec nos partenaires. Ce laboratoire est aussi le laboratoire de référence pour plusieurs projets, particulièrement le projet 'RAHYCO' , le projet « Seychelles » et « SkiPOGH » en collaboration avec le Dr. A.
