MALT Lymphomas and virus replications

A critical part of the replication cycle of many viruses is their interaction with the mucosal surface of the respiratory or gastrointestinal tract. This is the site of the initial interaction of most viruses and certain vaccines with their future hosts. The available data suggest that the biology of virus infections in polarised cells differs in several respects from that observed in the cell types commonly employed to study viruses.

The aim of this project is to study the interaction of human enteroviruses with polarised epithelial cells in particular the interaction of echovirus 11 with Caco 2 cells. Echovirus 11 interacts with a cell surface protein, decay accelerating factor (DAF/CD55), present on the apical surface of cells (ie the surface exposed to the external environment). In non-polarised cells this protein is necessary but not suffisant for virus entry. Echoviruses have the capacity to react rapidly to changes in their environment a fact that allowed us to derive mutant viruses that do not require this interaction with DAF/CD55 for entry.

Our studies to date reveal that binding of virus to DAF/CD55 results in actin dependent movement of virus in association with DAF/CD55 to tight juctions prior to internalisation of virus in vesicles. This movement causes a temporary reduction in tight juction function. We are in the process of studying the detailed mechanisms of this process.
