Parasitologie Moléculaire: La biologie d'un parasite intracellulaire obligatoire
Une des questions fondamentales de parasitologie consiste à comprendre comment une cellule eucaryote peut pénétrer, survivre et se multiplier dans une autre cellule eucaryote. La maîtrise de cette connaissance apparaît comme un pré requis à la mise en place de stratégies préventives et thérapeutiques des maladies infectieuse. Les membres de la famille des Apicomplexa ont une importance médicale et vétérinaire considérable, étant responsables d'une large gamme de maladies humaines et animales incluant la malaria, la toxoplasmose, les sporidioses et coccidoses. Parasites intracellulaires obligatoires, les Apicomplexa franchissent de façon active les barrières biologiques, pénètrent dans les cellules hôtes et en ressortent en utilisant un mode unique et conservé de motilité "glissée". Pour aborder l'étude de cette question complexe, nous avons choisi Toxopalsma gondii qui infecte presque tous les mammifères de même que les oiseaux, et qui colonisent quasiment toutes les cellules nucléées. C'est l'agent causal de la toxoplasmose, qui, habituellement, ne provoque pas de symptômes mais qui peut conduire à des complications sérieuses en cas d'infections congénitales et à une toxoplasmose cérébrale mortelle chez des individus immuno-compromis. Nous avons tout d'abord concentré nos efforts sur le développement de méthodologies qui permettent de manipuler génétiquement cet organisme. Plus récemment, nous avons établi un système inductible permettant l'étude de la fonction de gènes essentiels. impliqués dans la motilité, l'attachement à la cellule hôte, l'invasion et la survie intracellulaire.
