Mécanismes impliqués dans les propriétés de plasticité des synapses excitatrices en utilisant un modèle de cultures organotypique de cortex d'hippocampe.

L'objectif principal de nos recherches est de comprendre les mécanismes impliqués dans les propriétés de plasticité des synapses excitatrices en utilisant un modèle de cultures organotypique de cortex d'hippocampe. Les projets de recherche du laboratoire concerne les propriétés de la transmission synaptique, les mécanismes en affectant l'efficacité et la fonction, les événements moléculaires participant à la formation de nouvelles épines dendritiques, à la synaptogenèse et la croissance de neurites. Une question importante abordée par ces expériences est de comprendre comment l'activité neuronale contrôle et modifie l'organisation des réseaux synaptiques en situation physiologique mais aussi lors d'ischémie ou d'épilepsie. Ces questions sont étudiées à l'aide d'une approche multidisciplinaire associant des techniques électrophysiologiques (enregistrements intracellulaires par la méthode de patch clamp), de l'imagerie confocale 2-photons de cellules vivantes à l'intérieur d'un réseau complexe, de la microscopie électronique avec reconstruction tri-dimensionnelle de contacts synaptiques et des approches pharmacologiques et moléculaires.
