Radicaux libres et cellules souches embryonnaires
Dans notre groupe de recherche, nous travaillons sur deux thèmes principaux :
1. Radicaux libres - stress oxydatif - NADPH oxydase. 2. Cellules souches embryonnaires et différenciation neuronale. 1 : Les radicaux libres (aussi appelés formes réactives de l'oxygène) jouent un rôle clé dans le vieillissement et dans le développement des maladies liées au vieillissement. Ces radicaux libres ont également de nombreuses fonctions physiologiques cruciales allant de la défense contre l'infection à la régulation de l'expression des gènes en passant par la signalisation intracellulaire. Nous sommes particulièrement intéressés par les enzymes de la famille NOX, qui ont la fonction physiologique de produire des radicaux libres en transportant des électrons de NADPH vers l'oxygène. Les différents membres de cette famille d'enzymes sont exprimés dans pratiquement tous les tissus de notre organisme dans lesquels ils contribuent à des processus physiologiques mais aussi, lors de disfonctionnements, au développement de pathologies liées au stress oxydatif. 2 : Les cellules souches embryonnaires sont des cellules multipotentes représentant un potentiel très intéressant pour la science de base et pour les applications cliniques. Nous sommes particulièrement intéressés par la différenciation neuronale des cellules souches embryonnaires. Ceci dans le but de comprendre les mécanismes de la différenciation. Nous avançons vers le développement d'une thérapie cellulaire pour les maladies du système nerveux central. Nous tendons à un but particulièrement important à long terme : le développement de la thérapie cellulaire pour la maladie de Parkinson que nous tentons d'établir en réseau avec de multiples groupes des HUG.
