Métabolisme des lipoprotéines sériques et leur rôle dans le développement des lésions vasculaires
L'intérêt principal du laboratoire est le métabolisme des lipoprotéines sériques et leur rôle dans le développement des lésions vasculaires, notamment chez le patient diabétique. Les lipoprotéines
sont des complexes de protéines et de lipides dont la tâche principale est le transport et la redistribution des triglycérides, molécule primaire de stockage d'énergie, et du cholestérol, un composant
essentiel des cellules et le précurseur de nombreuses hormones. Les modifications au métabolisme des lipoprotéines perturbent le transport efficace des lipides et favorisent leur dépôt (surtout le
cholestérol) dans les parois des vaisseaux. Nos études portent sur les mécanismes par lesquels une sous-classe de lipoprotéines, les lipoprotéines de haute densité (HDL), protègent contre les lésions
vasculaires. HDL semblent agir par différentes voies, dont un est la réduction des conséquences du stress oxydatif. Ils empêchent ou limitent la peroxydation des lipides sanguins, et protègent également
l'endothélium contre le stress oxydatif. La capacité anti-oxydante des HDL est attribuée en grande partie à une enzyme associée, la paraoxonase (PON), qui est au centre de nos recherches. Les modèles
animaux ont clairement établi qu'elle empêche la formation de lésions vasculaires dues à son activité anti-oxydante. Un des projets est l'étude des facteurs qui influence l'expression du gène de la PON.
Nous avons rapporté des polymorphismes avec une forte influence sur les taux sériques de l'enzyme et, plus recemment, nous avons identifié des facteurs de transcription impliqués dans l'expression du
gène. D'autres projets ont étudié la voie par laquelle l'enzyme serait secrétée par le foie et s'associe aux HDL. Nous avons proposé un processus qui comporte l'insertion de la protéine dans la
membrane cellulaire avant son désorption par les HDL. Ce processus est de haute affinité, saturable et semble impliquer l'amarrage des HDL au surface des cellules. Complétant ces projets sont des études de
la PON dans un contexte plus clinique. Nous avons déjà démontré que les patients diabétiques sont déficients en paraoxonase et avons identifié d'autres facteurs environnementaux qui influençent son
capacité anti-oxydante, nottament le tabagisme. D'autres études de la fonction anti-inflammatoire de la PON, qui découle de son rôle anti-oxydant, sont entrepris dans un modèle transgènique de souris.
