Localisation et caractérisation des facteurs génétiques de maladies monogéniques et multifactorielles - Syndrome de Kartagener
Notre groupe, attaché à la section de recherche de la division de génétique médicale est spécialisé dans la localisation, l'identification et l'isolement des gènes responsables ou qui prédisposent à
des maladies monogéniques et multifactorielles chez l'humain.
Notre projet de recherche majeur porte sur l'identification des gènes responsables de la Dyskinésie Ciliaire Primaire ou syndrome de
Kartagener. Cette affection récessive se caractérise principalement par une motilité réduite ou absente des cils vibratiles des cellules épithéliales des voies respiratoires supérieures et du flagelle du
spermatozoïde. Ces anomalies ciliaires provoquent des symptômes respiratoires et une réduction de la fertilité masculine. L'hétérotaxie des organes (situs inversus) également observée dans 50% des
cas, est expliquée par une motilité anormale des cils qui contribue à l'établissement de gradients de molécules morphogènes chez le jeune embryon. Après avoir localisé plusieurs gènes de cette affection
génétiquement hétérogène, par analyse de liaison, nous caractérisons maintenant les gènes candidats et les analysons dans une cohorte de patients.
Nous avons également développé des outils et des
compétences dans les analyses de liaisons et d'associations, que nous mettons à disposition aussi dans le cadre de projets collaboratifs. Nous avons, dans un passé récent, réalisés des analyses de
liaison dans plusieurs maladies (psychiatriques, neurologiques, morphologiques, cancers).
