Diabete/Génétique/Transplantation d'îlots
Notre groupe étudie ces différents thèmes et cherche à définir les facteurs (nutrients,hormones,neuropeptides,...) impliqués dans la régulation des gènes codant pour les 2 hormones les plus
importantes dans le contrôle de l'homéostase du glucose, le mécanisme de leurs effets et les facteurs de transcription qui médient ces effets au niveau génique.
Dévelopement et différentiation des cellules à glucagon: Les mécanismes impliqués dans l'apparition des cellules à glucagon pendant le dévelopement et leur différentiation ne sont pas connus. Notre hypothèse est que ces cellules se différentient par un mécanisme de défaut, c'est-à-dire par l'absence de facteurs spécifiques aux autres cellules endocrines du pancreas, beta et delta ; en leur présence,la cellule à glucagon ne peut pas se développer, d'une part car ils favorisent l'expression des gènes spécifiques à ces cellules, comme l'insuline et la somatostatine et d'autre part, car ils inhibent l'expression du gène du glucagon. Nous testons ces hypothèses dans des lignées cellulaires et dans des animaux transgéniques. Bases moléculaires du diabète: Nous avons analysés les défauts génétiques impliqués dans la survenue du MODY(Maturity-Onset Diabetes of the Young) en Suisse, et d'agénésie de pancréas et de résistance sévère à l'insuline. Nous poursuivons ces analyses génétiques dans les pathologies humaines liées à des désordres de l'homéostase du glucose. Diagnostic moléculaire du rejet de greffe d'ilôts de Langerhans: Le diagnostic de rejet de la transplantation des ilôts de Langerhans est actuellement impossible. Nous avons mis au point un marqueur moléculaire de la perte d'ilôts après transplantation. Nous testons ce marqueur pour définir sa sensibilité et son utilité clinique.
