Fonction de l'îlot de Langerhans
Ce groupe étudie la fonction de l'îlot de Langerhans afin de mieux comprendre la défaillance fonctionnelle des îlots chez les patients diabétiques. Nous nous intéressons également à la thérapie
cellulaire du diabète Nous avons deux principaux projets:
1. La synthèse et la sécrétion de l'insuline:
La Proinsuline est acheminée vers l'aire de Golgi (TGN) où elle est triée afin d' être emballée dans
les granules sécrétoires. Par la suite, la proinsuline est transformée à l'intérieur des granules en insuline et C-peptide. Nous nous intéressons au mécanisme du tri dans le TGN ainsi qu'à celui de
la rétention sélective de l'insuline dans les granules. Nous étudions également les différentes étapes de l'exocytose et notamment le rôle des protéines SNARE et du cytosquelette. Ces études in vitro
nous aident à mieux comprendre les anomalies fonctionnnelles des cellules bêta dans les divers états pathologiques. Ils sont associés à l'dentification par "microarray" de gènes essentiels au bon
fonctionnement de cette cellule.
2. La sauvegarde de la masse des cellules bêta et le développement d'une thérapie
cellulaire du diabète
En collaboration avec le département de Chirurgie et des
groupes à Lausanne et à Zürich, nous aimerions élaborer une stratégie pour une thérapie cellulaire du diabète en remplaçant les cellules bêta défaillantes ou absentes par des cellules de remplacement.
Notre groupe cherche à améliorer la survie des cellules bêta humaines en culture. Ces cellules sont particulièrement sujettes à la mort par apoptose. Nous tentons de comprendre les voies de
signalisation de cette apoptose et de la prévenir, soit à l'aide d'inhibiteurs solubles, soit par l'expression de gènes appropriés. De telles interventions pourront également servir à préserver la masse de
cellules bêta chez l'homme dans des conditions d'apoptose élevée.
