Amnesia
This project explores the basis and mechanisms of amnesia and how the healthy human brain adapts thought to present reality. The clinical phenomenon at the basis of this research is spontaneous
confabulation, a profound misinterpretation of ongoing reality in thinking. The patients think and act according to memories which may have justly guided their behavior in the past, but which do not
pertain to ongoing reality.
Our recent results: 1) Thought adaptation to reality: We have discovered a mechanism, which depends on the posterior medial orbitofrontal cortex and which appears to be crucial for adjusting thought and behaviour to ongoing reality. It appears that this region adapts the electrocortical format of an upcoming memory according to its relation with ongoing reality. This adaptation occurs even before the precise content of a memory is recognized. We are currently testing the link of this capacity with the reward system and dopaminergic modulation. (2) Mechanisms of amnesia: using electrocortical brain mapping, we have discovered in an amnesic patient that he failed to normally activate distributed cortical networks. This result has now been confirmed in a group of amnesic subjects. We found item-specific efficacy and are currently exploring the limits (intensity) and the specificity of this treatment modality. Factors influencing spatial awareness and exploration in hemispatial neglect and rehabilitation of neglect (Program Director: Dr. R. Ptak).
