Signaux intracellulaires
Nous étudions le rôle du calcium et du pH sur les fonctions cellulaires, ainsi que les mécanismes contrôlant l¿activité de ces ions dans les différents compartiments de la cellule. Alors que le
maintien d¿un pH optimal est essentiel pour la plupart des réactions biologiques, des élévations locales de calcium peuvent activer des fonctions aussi diverses que la contraction musculaire ou la
transmission synaptique. Nous utilisons des protéines fluorescentes dérivées de la calmoduline pour mesurer les signaux calciques à l¿intérieur des organites intracellulaires. Ces sondes peuvent être
introduites, par des techniques de biologie moléculaire, soit dans le cytosol, soit dans le noyau, les mitochondries ou le réticulum endoplasmique.Cette approche est utilisée pour étudier le rôle de la
calréticuline, la principale protéine stockant le calcium à l¿intérieur du réticulum endoplasmique. A cette fin, nous mesurons les signaux calciques dans le cytosol et dans le réticulum de cellules dont le
niveau d¿expression de calréticuline
peut être modulé de façon spécifique. Le rôle du calcium nucléaire dans l¿activation de gènes est étudié à l¿aide de protéines fluorescentes, placées sous le
contrôle d¿un promoteur d¿intérêt. Une
concentration stable de calcium est établie dans le noyau par microinjection, et l¿activation de la transcription est mesurée par fluorescence.Nous étudions également
les mécanismes de régulation du pH intracellulaire. Le transport conductif de protons par la NADPH oxydase, une enzyme qui catalyse le transfert d¿électrons à travers la membrane plasmique, est
étudié par des techniques d¿électrophysiologie et de fluorescence.
