A) Notre groupe étudie les voies d'activation de l'immunité innée au niveau de la peau, un organe constamment exposé à des agressions microbiennes, chimiques et physiques. Nous concentrons notre recherche sur le rôle d'une nouvelle classe de senseurs du danger, qui représente l'équivalent intracellulaire des Toll récepteurs, et active des cytokines pro-inflammatoires comme l'IL-1beta. Nos travaux récents ont permis de mieux comprendre les mécanismes moléculaires permettant le développement d'une hypersensibilité de contact (eczéma) due à la pénétration de molécules sensibilisantes dans la peau. Grâce à des modèles murins et à des expériences in vitro, nous avons pu montré que les sensibilisants de contacts activent les senseurs du danger de type NOD-like receptors (NLR) au niveau du kértinocyte, et que cette activation est essentielle pour l'activation de cellules T effectrices. Nous étudions actuellement le rôle déterminant des NLR dans la sensibilisation et la tolérance aux antigènes en contact avec notre peau. Nous étudions également l'implication de mécanismes similaires dans la modulation de la carcinogenèse induite par les UV et le développement du psoriasis.

B) Un dysfonctionnement dans la voie de signalisation de l'Ectodysplasin A (EDA), un membre de la famille du TNF, entraîne une maladie sévère appelée dysplasie ectodermique hypohydrotique, caractérisée par l'absence des annexes de la peau que sont les poils, les dents et les glandes sudorales. Nous avons récemment produit une forme recombinante d'EDA qui permet de traiter cette maladie chez la souris (ODD acceptée à l'EMEA). Nous participons aux études pré-cliniques nécessaires au développement d'un traitement chez l'homme.
