Couplage cellulaire et connexines
Les connexines sont des protéines intégrales de membrane qui sont exprimées par pratiquement tous les types cellulaires et dans toutes les espèces animales. Leur structure primaire et leur
topographie ont été très conservées au cours de l'évolution. Chez les mammifères, ces protéines forment une famille multi-génique, comprenant une vingtaine d'iisoformes.
Les connexines sont concentrées dans des
régions membranaires qui établissent des contacts entre cellules adjacentes. A ces endroits, appelés jonctions " gap ", les connexines forment des canaux inter-cellulaires, au travers desquels les
cellules échangent des ions et des métabolites cytoplasmiques. Ce phénomène est appelé couplage cellulaire.
Les fonctions physiologiques du couplage cellulaire sont encore peu comprises. Cependant,
l'observation que des modifications de diverses connexines dans des animaux transgéniques résulte en de multiples et sévères phénotypes, indique que le couplage assure des rôles importants dans la
régulation du fonctionnement cellulaire. Plus récemment, la découverte que des mutations de connexines sont liées à diverses maladies humaines héréditaires, a indiqué que les connexines jouent également un
rôle pathogénique crucial dans certains organes, via un mécanisme encore non élucidé.
En utilisant des approches de biologie cellulaire et moléculaire, notre équipe étudie ce mécanisme de
signalisation et les fonctions de plusieurs connexines dans divers tissus épithéliaux et nerveux, in vitro et in vivo.
